Se non arriva nemmeno a 1:1 allora le cose diventano enormemente più semplici, niente slitta micrometrica e focus stacking ma semplicemente chiudi il diaframma tra f/11 e f/16 (fai un paio di prove), iso 100 e assicurati che il treppiede non si muova, scatta con un telecomando o con l'autoscatto ritardato.
Mi hai fregato 
Colpa mia, non ho cercato in rete e davo per scontato che, visto che si trattava di una colonia, fosse davvero un ingrandimento spinto, magari con tubi di prolunga.
