Se bene si voglia che questa antica basilica sia stata eretta nel secolo v, sopra le rovine di un tempio di Diana, da papa Gelasio (462-69), è certo che fu completamente rifatta dalle fondamenta nel 772 da Adriano I. Nel 1144 Lucio II la riunì a S. Giovanni in Laterano cui tuttora è sottoposta. Nel 1190 Celestino III la consacrò solennemente. Nel 1433 il Capitolo lateranense fece rifare il campanile che era rovinato e nel 1438 restaurò il portico ridotto in cattivo stato. Nel 1566 il cardinale Crivelli fece eseguire un importante restauro nella chiesa, restauro che fu proseguito nel 1570 dal cardinale Albani. Nel 1633 il cardinale Francesco Paolucci vi fece nuove aggiunte e nuovi restauri. Nel 1656 il Capitolo lateranense rinforzò il campanile che minacciava rovina e rifece porre una nuova campana. Nel 1702 il cardinale Sperello Sperelli vi fece eseguire alcuni lavori preservativi e finalmente negli ulti lui anni del secolo xviii il cardinale du Belby la riedificò parzialmente essendo molto trascurata e rovinosa. Portico. - La chiesa è preceduta da un portichetto, oggi mezzo chiuso. Interno. - È diviso in tre navi sorrette da 10 colonne di vari marmi tolte a edifici romani. Sul pavimento, tracce di formelle cosmatesche. Le pitture nelle pareti laterali della navata centrale sono di Paolo Gismondi detto Perugino. A destra: sepolcro del cardinale di Belloy arcivescovo di Parigi, morto nel 1808. Cappella in fondo all'abside: S. Antonio di Filippo Evangelisti. Altar maggiore: il paliotto è opera cosmatesca che rimonta alla fine del secolo xii. Il quadro a olio, rappresentante il santo titolare, fu eseguito nel 1570 da Federico Zuccari. Ai due lati, due affreschi di scene dell'Antico Testamento di Filippo Lauri. Nella sacrestia: pittura di Giovan Battista Brugi.
