La Cappella del Santissimo Sacramento si trova alla sinistra del presbiterio del Duomo di Spoleto. Realizzata alla fine del XVI secolo e decorata alla fine del XVIII, la cappella è caratterizzata da un altare marmoreo, mentre sulle pareti possiamo ammirare sei affreschi intervallati da statue di marmo. La cappella chiusa a seguito del terremoto del 1997, è stata riaperta al pubblico solo nel 2011 dopo un'opera di restauro che aveva interessato anche la Cappella della Santissima Icona.
