È situata lungo il lungarno Gambacorti, sulle rive del fiume Arno. La chiesa fu costruita nel 1230 e originariamente chiamata Santa Maria di Pontenovo per via del ponte vicino che collegava le due sponde. Il nome attuale deriva dalla reliquia di una spina della corona di Cristo, qui conservata dal 1333.
