Della Murcielago... la Lamborghini Automobili... iniziò la costruzione nel 2001 con la versione Coupè mentre nel 2005 vide la luce la versione Roadster. Fu costruita fino al 2010 in oltre 4000 esemplari. Nel 2006 la Roadster venne modificata e fra i vari aggiornamenti le venne aumentata la cilindrata del 12 cilindri a V portandola a 6496 cm3. Anche la potenza aumentò arrivando a 640 cavalli per una velocità massima di 340 chilometri orari. Il consumo dichiarato era di circa 4,6 chilometri con un litro di benzina contenuta nel serbatoio da 100 litri. La trazione era integrale sulle quattro ruote. Il nome e la sigla: Murcielago... cioè pipistrello nella lingua spagnola... fu il nome di un vigoroso toro vissuto nel 1879 mentre LP indica la posizione longitudinale posteriore del propulsore e 640 sono i cavalli da lui erogati. La vettura fu progettata dall'ingegnere e desiner belga Luc Donckerwolke.
