La costruzione del complesso monumentale di S. Chiara ebbe inizio nel 1310, per volontà del re Roberto d'Angiò e della sua seconda moglie Sancia di Maiorca. I lavori furono eseguiti sotto la direzione di Gagliardo Primario prima, e Lionardo di Vito poi. Nel 1340 la chiesa fu aperta al culto. La cittadella francescana fu realizzata costruendo due conventi contigui ma separati: uno femminile, destinato ad accogliere le clarisse, e l'altro maschile, ospitante i frati minori francescani. La chiesa si presenta oggi nelle sue originarie forme gotiche provenzali, con una facciata a larga cuspide, nella quale è incastonato l'antico rosone traforato, con il pronao dagli archi a sesto acuto. L'interno è a navata unica con dieci cappelle per lato. Caratterizza il presbiterio la presenza di monumenti funebri della famiglia reale angioina. Al centro campeggia la tomba di Roberto d'Angiò realizzata dai fratelli Bertini, mentre le due tombe sul lato destro, destinate ad accogliere le spoglie di Carlo di Calabria e Maria di Valois, si devono al grande maestro Tino di Camaino. La tomba sul lato sinistro, invece, è di Maria di Durazzo, realizzata da uno scultore anonimo, denominato appunto Maestro Durazzesco. Oltre ai monumenti funebri angioini in S. Chiara, nell'ultima cappella sulla destra, sono accolte anche le spoglie della famiglia dei Borbone. Nel 1742 la chiesa subì delle modifiche per opera dellíarchitetto Domenico Antonio Vaccaro. Fastosi rivestimenti donarono al complesso un aspetto barocco. Il 4 agosto del 1943 la chiesa venne quasi del tutto distrutta da un bombardamento aereo. Essa fu ricostruita e restaurata sotto la direzione di Mario Zampino, secondo l'originario stile gotico. Dieci anni dopo, il 4 agosto del 1953, la chiesa fu riaperta al culto. Alle spalle dell'altare maggiore è situato l'ex Coro delle clarisse, ambiente dal quale le monache partecipavano alle funzioni religiose. La cappella, strutturata come una sala capitolare cistercense, è composta da tre navate, due delle quali coperte da volte a crociera. Il Coro, oggi Cappella dell'Adorazione, è uno spazio riservato alla preghiera, pertanto è visitabile solo previa prenotazione.
