Di chiaro stile rinascimentale è la cupola che caratterizza il panorama lauretano, visibile in un vasto territorio che va dal mare alle valli collinari vicine. Il tamburo ottagonale è stato elevato fino al cornicione da Giuliano da Maiano, e compiuto nella calotta da Giuliano da Sangallo voltata in soli nove mesi, dal settembre 1499 "alle ore XV del 23 maggio" del 1500, come l'architetto annota nel suo diario. Era sabato e «... io Giuliano di Francesco di Sangallo fiorentino, con grandissima solennità e devozione e precisione, murai l'ultima pietra». La cupola, dal diametro di ben 22 metri, era inferiore all'epoca della sua costruzione, solo a quella brunelleschiana del Duomo di Firenze, a cui è palesemente ispirata.
