Inserita in uno dei quartieri in cui maggiormente pulsava la vita cittadina torinese, a pochi passi dall'Università, all'ombra della Torre del Comune che ancora svettava sulle case medievali di via Dora Grossa, l'edificio, che allora era la chiesa parrocchiale di San Gregorio, ricoprì sempre un ruolo importante riunendo intorno a sé un vasto pubblico di fedeli, composto per la maggior parte da ricchi borghesi e commercianti. Trasformatasi nella sede dell'Arciconfraternita di San Rocco, assunse un'importanza anche più grande nei momenti di calamità, nelle varie pestilenze che afflissero a più riprese la città, dove il suo compito di dar sepoltura ai cadaveri abbandonati diveniva quanto mai importante. Altare maggiore: progettato nel 1755 dall'architetto Bernardo Antonio Vittone, è composta da marmi policromi e reca sulla cimasa quattro statuette dorate intercalate da candelabri. Le statue rappresentano quattro dottori della Chiesa. Sotto la mensa è conservata l'urna con la reliquia di san Rocco. Presbiterio: Il pavimento è un intarsio di lastroni di marmo variamente colorati; la balaustra risale al 1745 ed è opera di Giovanni Batista Morari (fine sec. XVIII). Nella nicchia absidale campeggia la tatua lignea del santo scolpita da Pietro Botto, collocata inizialmente sulla facciata esterna (sec. XVII). Bellissimi gli stalli del coro scolpèiti nel 1722 da Francesco Gilardi e Giuseppe Maria Bonzanigo. L'affresco dela volta rappresenta episodi della vita del Santo e si deve al pittore di origine luganese Rocco Comaneddi (1791). Alle pareti laterali due dipinti;: uno di Giovanni Antonio Mari con la Predica di san Rocco agli appestati, l'altro con la Morte del Santo di Tarquinio Grassi (II metà sec. XVII).
