Solo nel 1634 si pone solennemente la prima pietra per edificare una nuova chiesa, su disegni del Vitozzi e/o del Castellamonte. I lavori non proseguono oltre le fondamenta. In questo periodo arrivano a Torino i padri Teatini, ordine sacerdotale dedito allo studio, alla catechesi, all'insegnamento delle scienze.Guarino Guarini, padre teatino, architetto, è invitato a Torino dal duca Carlo Emanuele II nel 1666. Nato a Modena nel 1624, entra a 15 anni nella congregazione dei preti di S. Gaetano, fondatore dell'ordine, insegna lettere e filosofia a Messina, viaggia in vari paesi dell'Europa tra cui Lisbona e Parigi per seguire i cantieri dei suoi progetti e studiare l'architettura antica. Nel 1666 riceve dal duca sabaudo l'incarico di edificare una nuova chiesa dedicata a s. Lorenzo. L'inaugurazione della stessa avviene il 12 maggio 1680: poco più di 13 anni per costruire la complessa struttura. Chiuso il lanternino, Guarino Guarini celebra la santa Messa alla presenza di tutta la corte Sabauda (come documentato negli archivi di stato); viene cantato il Te Deum di ringraziamento, musicato per l'occasione dal maestro Giovan Battista Carisio.L'arditezza della costruzione è degna di un grande genio: durante la seconda guerra mondiale una bomba ha sventrato il palazzo situato a fianco della chiesa, oggi palazzo della Regione; malgrado le forti vibrazioni, la struttura non ha subito danni. La Real Chiesa fu tutta affrescata per ordine del re Carlo Felice nel 1827 dai fratelli Fea di Casale e restaurata nel 1998 in occasione dell'ostensione della Sindone.
