La chiesa del Santo Sudario nacque per volontà della Confraternita del Santo Sudario di Torino, fondata nel 1598 e attiva tutt'oggi. Nel 1728 Vittorio Amedeo II di Savoia concedeva alla Confraternita l'area occupata attualmente dal vecchio ospedale per i malati di mente (spedale dei Pazzerelli), anticamente nota come Isolato di Sant'Isidoro; qualche anno più tardi, nello stesso lotto, veniva eretta una cappella per le funzioni religiose dell'ospedale. Il progetto della chiesa venne affidato a Ignazio Mazzone, architetto e confratello, mentre la scelta dei pittori per decorarne le pareti interne venne affidata a Claudio Francesco Beaumont: verranno scelti i nomi di Michele Antonio Milocco per il trompe-l'?il, aiutato dal veneziano Pietro Alzeri. La facciata, originalmente non prevista nel progetto, venne affidata a Giovanni Battista Borra, e la chiesa venne aperta al pubblico nel 1764. Attualmente l'accesso alla chiesa è inserito all'interno del percorso di visita del Museo della Sindone, ospitato nella cripta dell'edificio. Particolare interessante sulla volta, la bandiera italiana nella rappresentazione degli Angeli che sostengono il Sudario, sicuramente un'aggiunta voluta dai Savoia al momento dell'Unità d'Italia.
