Il Medagliere deriva dal Gabinetto delle Medaglie del re Carlo Alberto che acquista, nel 1832, la collezione di monete antiche e medioevali di Domenico Promis, nominato contestualmente conservatore del Gabinetto.Tra il 1835 e il 1838, infatti, il re fa riallestire appositamente un ambiente annesso alla Galleria Beaumont dall'architetto Pelagio Palagi; la decorazione della sala, che una porta collega al Gabinetto Cinese di Palazzo Reale, è stata ideata dal progettista per ospitare la collezione di monete, di medaglie e di sigilli raccolta da Carlo Alberto: a questo scopo sono stati progettati gli arredi intarsiati ed eseguiti dall'ebanista Gabriele Capello. Le vetrine e le cassettiere richiamano, nella loro forma e nelle loro decorazioni, i templi antichi, mentre i tavoli sono stati pensati per facilitare la consultazione della raccolta. Il fregio in alto è stato dipinto da Pietro Ayres con putti che reggono ghirlande fiorite e medaglie, con un'evidente allusione agli oggetti conservati nella sala: ne è l'autore Pietro Ayres da Savigliano che, con Diego Marielloni, ha eseguito anche i dipinti del soffitto con figure mitologiche e allegoriche.Nelle vetrine sono inoltre esposti alcuni oggetti d'arte scelti dal sovrano dalle raccolte di oggetti preziosi dei Savoia per arricchire la sala. Attraverso acquisti e doni la raccolta di monete, medaglie e sigilli la collezione attuale annovera circa trentatremila pezzi.
