L'attuale edificio di Santa Maria di Castello è una basilica romanica a tre navate con transetto e tre absidi edificata nel primo quarto del XII secolo ad opera di maestranze antelamiche. L'aspetto primitivo della chiesa è stato in parte alterato dalle ristrutturazioni dei secoli XV e XVI. Dalla Cappella di S.Tommaso si accede alla grande cappella del Crocifisso, in cui è collocata la copia del Cristo moro (l'originale in legno del XIII sec.) nella versione barocca. Nella zona di accesso, affresco raffigurante la Vergine addolorata, di Gregorio De Ferrari e i monumenti funebri di due arcivescovi di Genova, appartenenti all'ordine domenicano: Giulio Vincenzo Gentile e Nicolò Maria De Franchi, opera rispettivamente di Filippo Parodi e Pasquale Bocciardo.
