Nell'omonima piazza sorge il Duomo, uno degli esempi più insigni di cattedrale romanico-gotica italiana. Secondo gli studi il Duomo sostituisce una prima chiesa dedicata a Maria, eretta intorno al sec. IX, sorta nel luogo in cui si trovava un tempio offerto a Minerva. La Cattedrale conserva numerosi capolavori e l'opera principale è il pavimento. Iniziato dal XIV secolo, è stato concluso soltanto nell'Ottocento dai più importanti pittori e scultori, tutti senesi, tranne Bernardino di Betto detto il Pinturicchio, autore, nel 1505, della tarsia con il Colle della Sapienza. Nel Duomo hanno lavorato i più grandi scultori di ogni epoca: da Nicola Pisano, che realizzò il pulpito dal 1265 al 1268, al figlio Giovanni, autore delle sculture della facciata, da Donatello, di cui resta la statua del Battista nella omonima cappella, a Michelangelo, che scolpì San Pietro e San Paolo, San Pio e Sant'Agostino per l'altare Piccolomini, fino Gian Lorenzo Bernini, della cui arte recano testimonianza la Maria Maddalena e il San Girolamo della Cappella del Voto. Nella cattedrale di Siena restano anche numerose pitture: tavole, tele e cicli a fresco. La Libreria Piccolomini costituisce un unicum, un monumento di assoluto rilievo per il ciclo di affreschi, eseguiti da Pinturicchio e dalla sua bottega, fra il 1503 e il 1508. Nella zona presbiteriale, si osservano i quattro grandi affreschi di Ventura Salimbeni (1610), il pittore che, insieme al fratellastro Francesco Vanni e ad Alessandro Casolani, è protagonista dell'arte senese tra Cinque e Seicento. Di notevole interesse, infine, la serie delle pale degli altari del Duomo, ancora oggi poco conosciute, e sono opere assai rappresentative del XVII . La Cattedrale conserva inoltre una serie di pregevoli vetrate. La più importante, quella di Duccio di Buoninsegna, un tempo collocata sopra l'abside, è stata di recente trasferita nel Museo dell'Opera e sostituita da una copia. Nell'occhio della facciata è invece posta una vetrata raffigurante l'Ultima cena, eseguita nel 1549 da Pastorino de' Pastorini, allievo di Guillaume de Marcillat. Ricordiamo infine il coro, una delle opere più conosciute della lunga e feconda carriera artistica di Fra Giovanni da Verona (1456-1525), intarsiatore e intagliatore di grande rilievo nel panorama storico-artistico del XVI secolo.
