"Erano i Giganti grandi di statura, che da' lampi de' fólgori percossi ruinavano a terra... "[G. Vasari] L'ambiente, eseguito in via definitiva tra il 1532 e il 1535, narra la vicenda della Caduta dei Giganti, tratta dalle Metamorfosi di Ovidio. La camera è la più famosa e spettacolare del palazzo, sia per il dinamismo e la potenza espressiva delle enormi e tumultuose immagini, sia per l'audace ideazione pittorica, volta a negare i limiti architettonici dell'ambiente, in maniera tale che la pittura non abbia altri vincoli spaziali se non quelli generati dalla realtà dipinta. Giulio Romano infatti interviene per celare gli stacchi tra i piani orizzontale e verticale: smussa gli angoli tra le pareti, gli angoli tra le pareti e la volta e realizza un pavimento, oggi perduto, costituito da un mosaico di ciottoli di fiume che prosegue, dipinto, alla base delle pareti. Con questo stupefacente artificio unitario e illusionistico, l'artista intende catapultare lo spettatore nel vivo dell'evento in atto, per produrre in lui stupore e sensazione di straniamento.
