La chiesa della Santissima Vergine Spiliotissa, chiamata anche Mitropolis dal nome della piazza in cui sorge, è situata di fronte al porto vecchio. La chiesa, rialzata rispetto alla piazza e con una scalinata, risalta subito per il suo colore accesso e per la decorazione che ricorda un sole intorno al rosone. Il nome Spiliotissa deriva da "spilià" che vuol dire grotta ed è il nome di questo quartiere chiamato così per la presenza di una grotta ai piedi della Fortezza Nuova. Questa basilica ortodossa risale al 1577 ed ospita le spoglie di Santa Teodora di Augusta, un'imperatrice bizantina canonizzata nel IX secolo, le cui reliquie sono arrivate a Corfu da Costantinopoli insieme a quelle del Santo Patrono San Spiridone. All'interno della chiesa ci sono icone bizantine come quella della Madonna Dimosiana, dipinta da entrambi i lati, e altre della Scuola Ionica ad opera di Damaskino, Tzane e Paramithioti.
