Edificio unico a forma conica del XVIII secolo che ospita un museo del patrimonio culturale con manufatti e arredi originali. Trullo Sovrano è situato nella parte nord di Alberobello, alle spalle della Chiesa dei Santi Medici Cosma e Damiano. Esso rappresenta l'unico trullo ad avere un piano sopraelevato, raggiungibile dall'interno attraverso una scala in muratura, tra i primi ad essere costruito con la malta. La maestosa cupola conica, alta circa 14 metri, si erge imponente al centro di un gruppo costituito da dodici coni. L'attuale ala sinistra costituisce il suo nucleo originario, che si può far risalire agli inizi del 1600, mentre la parte restante fu costruita nella prima metà del 1700 per conto della famiglia benestante del sacerdote Cataldo Perta (1744 – 1809). Nel tempo l'edificio ha avuto diverse destinazioni: corte, cappella, spezieria, cenobio, oratorio campestre, abitazione . Nei primi anni del 1800 vi dimorarono le reliquie dei Santi Cosma e Damiano – Patroni di Alberobello – e dal 1826 fu sede della Confraternita del SS.Sacramento. Sul finire del XIX sec. divenne di proprietà della Famiglia Sumerano (tuttora proprietaria), che fece del Trullo la propria abitazione. Gli arredamenti e tutti gli oggetti esposti sono autentici. Aprile 2015, è stato inserito nella lista dei siti di particolare interesse storico e artistico.
