Grazie agli studi compiuti da un gruppo di ricercatori internazionali pubblicato dalla prestigiosa rivista scientifica Molecular Biology Evolution, ora sappiamo che la Sardegna ha accolto genti arrivate dall'Oriente e dall'Occidente e che questo è avvenuto prima del Neolitico, nel Mesolitico, subito dopo l'epoca glaciale. Gli archeologi avevano già formulato tale ipotesi e le ultime ricerche confermano i loro studi come già aveva fatto la ricerca condotta da Paolo Francalacci e pubblicata su Science nel 2013. Il popolo sardo ha, quindi, nel suo patrimonio genetico caratteri sia orientali che occidentali. Ma ciò che appare ancora più interessante e che questa eredità si è conservata per millenni fino a giungere a noi con alcune caratteristiche immutate rendendo il DNA dei sardi unico. La ricostruzione di questo è stata possibile grazie a studi sul DNA antico che, con l'ausilio della biologia molecolare e della genetica evoluzionistica, ha permesso di accertare le origini dei sardi. La ricerca è stata condotta comparando il DNA dei Sardi attuali con quello dei sardi antichi ed anche con genomi di uomini moderni e antichi non sardi. Il lavoro ha richiesto l'analisi di un numero grandissimo di dati: 3491 campioni di DNA appartenenti a sardi moderni, 21 appartenenti a sardi antichi ( DNA estratto da campioni raccolti in siti archeologici datati da 4 a 6 mila anni fa), comparati con 50.000 genomi di uomini moderni e 500 antichi, tra cui vi era anche il DNA appartenente ad Ötzi. I risultati ottenuti da questo importante lavoro ci rivelano che la popolazione Sarda ha un patrimonio genetico unico con delle caratteristiche non riscontrabili in altre popolazioni europee. I risultati evidenziano che la Sardegna rappresenta un'anomalia nel panorama genetico europeo, e per più di un motivo. I ricercatori hanno scoperto che nel DNA dei sardi vi è una percentuale che va dal 3 al 5 per cento di eredità genetica lasciata da popoli che hanno abitato in l'Isola e vi siano giunti circa 8000/10.000 anni fa in un periodo antecedente alla introduzione dell'agricoltura in Sardegna. Si ipotizza anche che vi siano nel genoma dei sardi caratteristiche uniche ed appartenenti ai primi abitanti dell'isola residenti prima del Neolitico. Inoltre il 78,4 per cento del DNA estratto dai mitocondri dei sardi moderni appartiene a rami genetici che non si riscontrano in altre zone geografiche europee ma sono unicamente isolani. Si sono potuti classificare i genomi mitocondriali dei sardi in 89 gruppi genetici (aplogruppi) e mettere in evidenza caratteristiche genetiche, risalenti al neolitico che si sono conservate attraversando il periodi nel nuragico, post nuragico fino ad essere presenti ancora ora.
