Commissionata inizialmente dalla confraternita della Santa Croce, la chiesa venne eletta da Vittorio Amedeo II (1729) Basilica Magistrale della Sacra Religione dell'Ordine dei Santi Maurizio. Di gusto tipicamente barocco, la basilica è posta a ridosso della retrostante Galleria Umberto I e si sviluppa su una pianta centrale, la cui navata è interamente sormontata da un ottimo esempio di cupola su base ellittica, di influenza guariniana.La costruzione risale al 1740-99 ad opera dell'architetto luganese Antonio Bettino, che aveva tenuto presente i modelli inaugurati a Torino dal Lanfranchi. Nella soluzione concentrata dello spazio, definito da colonne rastremate e scanalate, emerge l'altare maggiore di Antonio Bertola, concluso tra il 1704; opere di Ignazio Perucca le statue lignee barocche con i dottori della Chiesa che affiancano l'altare maggiore. La facciata neoclassica è di Carlo Bernardo Mosca, 1834, ornata con le statue dei Santi Maurizio e Lazzaro realizzate da Giovanni Albertoni e Silvestro Simonetta. I bombardamenti del 13 luglio del 1943 che causarono ingenti danni a tutto il circondario danneggiarono gravemente il ciclo di affreschi ottocenteschi del Morgari e del Gonin. Contestualmente alle altre proprietà dell'Ordine Mauriziano, nel 2001 furono avviati dei lavori di restauro.
