La Ventesima Cappella è dedicata alla cerimonia della beatificazione dei San Francesco ed è senz'altro la più scenografica. A semicerchio decine di personaggi – opera di Dionigi Bussola – assistono alla consegna da parte di papa Gregorio IX della bolla di canonizzazione di San Francesco al Padre Generale dell'ordine. Re, aristocratici, ambasciatori, cardinali, vescovi, frati e figuranti popolano la cappella. Gli affreschi sono di Antonio Busca (Milano, 1625 – 1686).
