I numerosi reperti archeologici, le necropoli e le misteriose ed affascinanti Vie Cave ci testimoniano che la città sia stata abitata fin dall'età arcaica. Di origine senza dubbio etrusca, vi succedettero prima i romani e successivamente i Longobardi. Sin dal IX sec. a.C. la vita politica di questo paese fu strettamente legata a Sovana, sede principale della famiglia Aldobrandeschi. Successivamente con il declino inesorabile che stava conoscendo Sovana, l'importanza di Pitigliano divenne sempre maggiore e nel 1293, con il matrimonio tra Romano Orsini ed Anastasia, figlia di Margherita Aldobrandeschi e di Guido di Montfort, la sede della contea fu trasferita da Sovana a Pitigliano, ritenuta evidentemente più sicura. Alla morte del più illustre esponente della famiglia Orsini, papa Nicolò III, l'intera contea fu travagliata da conflitti interni e dagli assalti della repubblica senese. Il 26 maggio del 1546 Pitigliano fu teatro di un duello che all'epoca dei fatti fece grande clamore: si sfidarono i due noti capitani Giovanni Taddei nobile fiorentino ed Ascanio Della Corgna signore di Castiglione del Lago; per l'occasione giunsero a Pitigliano tremila tra cavalieri ed esponenti della nobiltà. Giovanni Taddei restò ucciso e questo duello fu uno degli ultimi duelli “autorizzati” infatti nel 1563 il Concilio di Trento proibì questa forma di controversia che tuttavia continuò ad essere praticata ancora per molto tempo ma in modo clandestino. Nel 1547 i cittadini di Pitigliano proclamarono signore delle loro città Niccolò IV, ma il suo governo durò poco e finì male. Il tribunale dell'Inquisizione pontificia infatti lo sottopose a giudizio e lo imprigionò. Dopo alterne vicende ed intrighi politici, nel 1604 Cosimo de' Medici prese possesso della città ma il dominio mediceo non portò gli effetti sperati ed il progressivo impoverimento cessò soltanto con l'avvento dei Lorena. Di qui in poi le sorti del paese seguirono quelle del Granducato di Toscana. Infine con il plebiscito del 1860, Pitigliano aderì al Regno d'Italia seguendone le vicissitudini. Pitigliano è anche conosciuta come la piccola Gerusalemme d'Italia per via di una comunità ebraica che vi arrivò nel XV sec. e che nonostante emigrazioni verso Firenze e Livorno si mantiene, seppur piccola, tutt'oggi.
